FP891 : Evaluation of Drug Reservoir Function of Human Amniotic Membrane (HAM) using a model drug – Prednisolone

Purpose:
Prednisolone is used for the treatment of corneal graft rejection and ocular surface disorders.Purpose of study was to investigate the reservoir function of Human Amniotic Membrane(HAM) in vitro using prednisolone phosphate.

Methods:
HAM buttons were punched using 12 mm trephine and categorized into thick and thin HAM based on wet weight.Buttons were soaked in freshly prepared topical prednisolone phosphate(1% w/v)solution,incubated for different time intervals.Release kinetics from drug soaked HAM was studied for 3 weeks using High Performance Liquid Chromatography (HPLC).

Results:
No significant difference was observed in release kinetics with different incubation time periods except 12 Vs 48 and 24 Vs 48 time periods (P value 0.009). Thick HAM showed significant increase in drug entrapment as compared to thin HAM (23 % Vs 11 %; p< 0.001)

Conclusion:
Present study showed that HAM may be a suitable drug reservoir for the treatment of corneal disorders in a sustained manner.
